Nous \u00e9tudions notamment\u00a0<strong><em>Vibrio cholerae<\/em><\/strong>, l&#8217;agent causal du chol\u00e9ra, et\u00a0<strong><em>Escherichia coli<\/em><\/strong>, y compris les souches cliniques responsables d&#8217;infections urinaires.<br>L&#8217;\u00e9mergence de la r\u00e9sistance aux antibiotiques peut \u00eatre associ\u00e9e \u00e0 la pr\u00e9sence de faibles concentrations d&#8217;antibiotiques dans les environnements aquatiques, comme les lacs et les rivi\u00e8res situ\u00e9s \u00e0 proximit\u00e9 des h\u00f4pitaux, des industries pharmaceutiques et des fermes d&#8217;aquaculture, o\u00f9 ces substances sont r\u00e9guli\u00e8rement utilis\u00e9es. Ces milieux abritent une grande diversit\u00e9 bact\u00e9rienne, dont des esp\u00e8ces de\u00a0<strong>Vibrio<\/strong>. De faibles concentrations d&#8217;antibiotiques peuvent \u00e9galement \u00eatre retrouv\u00e9es dans diff\u00e9rents compartiments du corps lors d&#8217;un traitement, notamment dans les poumons, o\u00f9 leur diffusion varie selon les conditions physiologiques, ou dans l&#8217;urine en fin de traitement.<br>La recherche sur la r\u00e9sistance aux antibiotiques se concentre g\u00e9n\u00e9ralement sur les m\u00e9canismes permettant aux bact\u00e9ries de survivre \u00e0 des doses \u00e9lev\u00e9es. En \u00e9tudiant l&#8217;effet des faibles doses, nous avons la possibilit\u00e9 d&#8217;identifier de nouvelles cibles impliqu\u00e9es dans l&#8217;acquisition de la r\u00e9sistance. Un m\u00e9canisme r\u00e9cemment identifi\u00e9 dans la tol\u00e9rance aux antibiotiques repose sur des modifications de l&#8217;ARN bact\u00e9rien.<br>L&#8217;ARN joue un r\u00f4le central dans la synth\u00e8se des prot\u00e9ines, \u00e0 la fois comme matrice et comme composant des machines cellulaires. Ses bases peuvent subir diverses modifications chimiques, comme la m\u00e9thylation, la pseudouridylation, la dihydrouridylation, l&#8217;incorporation de queuosine et d&#8217;autres alt\u00e9rations plus complexes. Ces modifications n&#8217;induisent pas toujours directement de r\u00e9sistance, mais influencent la tol\u00e9rance bact\u00e9rienne aux antibiotiques, ce qui peut compromettre l&#8217;efficacit\u00e9 des traitements et favoriser l&#8217;\u00e9mergence de r\u00e9sistances.<br>Notre projet s&#8217;int\u00e9resse \u00e0 un ph\u00e9nom\u00e8ne encore peu explor\u00e9 : la\u00a0<strong>&#8220;r\u00e9sistance ph\u00e9notypique&#8221;<\/strong>, o\u00f9 les bact\u00e9ries d\u00e9veloppent une tol\u00e9rance aux antibiotiques et finissent par devenir r\u00e9sistantes sans qu&#8217;aucune mutation g\u00e9n\u00e9tique connue ne soit d\u00e9tect\u00e9e dans leur g\u00e9nome. Pour comprendre les m\u00e9canismes sous-jacents, nous combinons biologie mol\u00e9culaire, \u00e9tudes g\u00e9n\u00e9tiques, analyses biochimiques et approches \u00e0 haut d\u00e9bit.<br>L&#8217;extraordinaire diversit\u00e9 des modifications de l&#8217;ARN, leur impact sur les caract\u00e9ristiques et comportements bact\u00e9riens, ainsi que leur sensibilit\u00e9 aux conditions environnementales, en font un champ d&#8217;\u00e9tude prometteur.
